La to Chile via public transport - Is it possible?

You cannot go to South America by bus or train. there are no trains, and there is no complete road...there is an area in an area in Panama called the Darien Gap that is impassable. As other guy said, you can fly to Ecuador, then take buss...would be quite an adventure.
